Overview

Ordering for MGH laboratories and staff (internal) is done through the Galaxy server, while external customers may submit samples by completing a spreadsheet and sending it to our core via email. For internal and external customers, we have specific guidelines which we ask that you follow in order to insure that your results are the best possible quality (see links below for more information).

 

Note: You must be registered as a customer before you may submit samples for sequencing or library construction services. MGH customers can register by clicking on Lab>Register in the top menu bar of the Galaxy server. All other customers may register by filling out the New Customer information sheet (see below) and emailing it to nextgen@research.mgh.harvard.edu.

All Users

To make the submission process go more smoothly and to ensure you receive the highest quality data, we provide a sample submission checklist that we recommend all customers to follow.

 

Failure to complete the checklist items may result in the return of your sample unprocessed. If you cannot meet a particular requirement, please contact us to work out a solution.

MGH (Internal) Users

To register as a customer and submit a sample, please go to the Galaxy server, register, and complete the online submission form.

Make sure your samples follow our submission guidelines and are delivered to the correct room (refer to Policies below).

Non-MGH (External) Users

To register as a customer, please email sequencing@molbio.mgh.harvard.edu to tell us about yourself and indicate your interest in sequencing with us. We will let you know what billing information we need from you.

 

To submit a sample, please download the Sample Submission Form. For best results and to save the document, do not open in Google Chrome.

 

Fill in completely and email to nextgen@research.mgh.harvard.edu. For returning customers, the customer information listed at the top of the submission form is sufficient.

Sample Submission Policies

 

There are requirements for the library construction and validation processes which, in some cases, must be addressed before the final library is complete. PLEASE READ ALL REQUIREMENTS BEFORE STARTING LIBRARY CONSTRUCTION.

QC/Validation

The Sequencing Core strongly recommends that all libraries be validated before submission to the lab.

 

The following are recommended QC methods:

• qPCR

• PicoGreen

• Bioanalyzer

 

*Note: We do not recommend using Nanodrop as a validation method. Nanodrop tends to significantly overestimate the amount of material that will be viable on the flowcell.

 

Submission

We also strongly recommend that samples be diluted to 10-20nM, with a maximum submitted concentration of 100nM, if necessary. We request that samples be submitted at a volume no less than 10µl whenever possible (although more is better) and in low-bind tubes (e.g. Fisher brand siliconized low-retention tubes, cat#02-681-320).

 

Galaxy's online form will automatically generate a sample ID for each of your samples, and all samples must be labeled with this sample ID. The downloadable submission form provides instruction on assigning a sample ID for external customers.

 

Custom Primers

Custom sequencing primers may be submitted for sequencing special libraries or libraries with an initial constant region (e.g. the fragments contain a GATC sequence at the beginning of the insert). It is important to remember that the Illumina platform only allows for custom sequencing primers for READ 1 of sequencing!

 

The submitted primer tube must have the following characteristics:

  • HPLC purified
  • Reconstituted to 100uM in DI water, EB, etc. (we recommend a standard SSC buffer with TWEEN)
  • 10ul volume
  • In a low-bind tube
This is IMPORTANT!

There is no extra charge for using a custom sequencing primer.

 

Sample Delivery

Our facility is located in the Simches Research Building (MCPZN) at 185 Cambridge St. MGH customers may deliver samples to Ulandt Kim (tel. 617-643-0745) or placed in the freezer as instructed by him. You can find directions to the Simches Research building here.
